Scientists from the University of Exeter, working with global research partners, have discovered a previously unidentified form of neonatal diabetes caused by mutations in a single gene that prevents insulin-producing cells from functioning properly. The condition, known as monogenic neonatal diabetes, is inherited in the majority of cases.
According to researchers, mutations in the TMEM167A gene interfere with the body’s ability to produce insulin, leading to dangerously high blood sugar levels in infants. In some cases, the disorder is also associated with neurological conditions such as epilepsy and reduced brain development.

Dr. Elisa De Franco from the University of Exeter explained that identifying the genetic roots of diabetes in newborns offers critical insight into how insulin is produced and secreted in the body. “These discoveries help us understand both rare and more common forms of diabetes,” she said.
The study examined children who showed both metabolic and neurological symptoms, all of whom shared mutations in the same gene. Laboratory experiments confirmed that damage to this gene causes insulin-producing cells to fail and eventually die due to internal stress.
Health professionals say early warning signs in newborns may include poor weight gain, frequent urination, dehydration, and unusually high blood sugar levels. In severe cases, dangerous ketone buildup may also occur.
