Key Developments:

  • Blood flow activates signals that help define liver cell functions
  • Findings could inform future research into liver disease and cancer

(Medical Xpress) — October 12, 2026: The way blood moves through the liver may help determine how its cells specialise, according to research published in Nature Communications. Scientists have identified a mechanism through which physical forces associated with circulation influence chemical messages between blood vessels and surrounding liver tissue.

The investigation, conducted by researchers at Heidelberg University’s Medical Faculty Mannheim and the German Cancer Research Center, focused on the relationship between the liver’s vascular network and its metabolic organisation. Experiments in genetically modified mice, combined with single-cell and spatial molecular analyses, helped the team examine how blood vessels respond to their physical environment.

The liver is responsible for processing nutrients, storing energy and breaking down substances, among other essential tasks. These responsibilities are distributed across different areas rather than performed identically by every liver cell. Scientists call this arrangement metabolic zonation: the position of a cell within a liver lobule helps determine which biological processes it carries out.

The research examined how this regional specialisation is maintained. Cells lining the liver’s smallest blood vessels release biochemical messengers that influence nearby hepatocytes, the cells responsible for many of the organ’s metabolic functions. These messengers include Wnt signalling molecules, which help regulate cellular behaviour and identity.

The researchers found that the production of these signals is influenced by the mechanical forces generated as blood travels through the liver. Endothelial cells respond to these forces by activating molecular programmes associated with particular regions of the vascular network. The resulting Wnt signals help establish and maintain the specialised functions of neighbouring liver cells.

This suggests that tissue organisation depends not only on chemical conditions, such as oxygen availability, but also on the physical environment surrounding cells. According to the study, the distribution of blood-flow-related forces helps explain why different vascular regions produce distinct signalling patterns.

Senior author Hellmut Augustin highlighted the implications for understanding the relationship between circulation and tissue function. He said: "Blood vessels are not passive pipelines. They actively measure mechanical forces generated by blood flow and translate them into molecular instructions that organize the surrounding tissue."

The investigation also revealed a coordinated arrangement within the vascular network itself. Particular Wnt receptors were concentrated in locations where the corresponding signalling molecules were produced, creating local communication systems. These arrangements appear to help preserve the molecular characteristics of different vascular regions and support the organisation of nearby liver tissue.

First author Dr Ki Hong Lee described the role of endothelial cells in translating physical conditions into biological responses. He said: "They read physical information from blood flow and convert it into biochemical signals that tell neighboring liver cells which functions they should perform."

The effects were not confined to the cells surrounding blood vessels. The same signalling mechanisms also influenced endothelial cell structure and the molecular connections these cells form with their vascular neighbours. The findings therefore point to a system in which blood-flow forces help shape both the vessel lining and the tissue it supports.

The work may help researchers investigate conditions in which the liver’s normal organisation becomes disrupted. Changes in metabolic zonation are relevant to the study of chronic liver disease, fibrosis and liver cancer. Understanding the signals that maintain healthy tissue structure could eventually help scientists explore new approaches to treating such disorders.

However, the findings represent a step in understanding liver biology rather than a new treatment. The study relied substantially on mouse experiments, and further research is needed to establish whether the same mechanisms operate in humans in comparable ways. Scientists must also determine whether modifying these signals could safely improve outcomes in people with liver disease.

The study, titled Haemodynamic control of zonated liver function via instructive vascular Wnt signalling, provides evidence that the forces created by circulation can influence the molecular communication responsible for organising liver activity. Its findings broaden scientific understanding of how physical conditions within an organ can help determine the functions of its cells.
